Tottenham had made the signing of a new right wing-back one of their priorities for the summer transfer window.
The deal has now been made official and Spence becomes Tottenham's sixth signing of the summer.
Spurs have already bought Richarlison for £60m from Everton and Yves Bissouma from Brighton for £25m, plus the free transfers of Ivan Perisic and Fraser Forster, and the loan of Clement Llenglet from Barcelona.
This signing of Spence, 21, takes Tottenham's summer spending to close to the £100m mark.
Spence had generated interest from as many as 10 European clubs, with Bayern Munich even sending scouts to the City Ground to watch him last season.
The youngster played a pivotal role as Forest were promoted to the Premier League for the first time in over two decades.
He made 46 appearances in all competitions last term, scoring three goals.
His strengths are primarily offensive, where he shines with his eye-catching dribbling, but he also rates well in terms of recoveries and defensive positioning.
